The Construction of Molecular Linkage Frame Map in Chinese fir

Abstract: Chinese fir ( Cunninghamia lanceolata ( Lamb.) Hook) is a fast growing evergreen coniferous tree species with wide distribution areas which cover all the subtropical in southern China. Research Group at Nanjing Forestry University made the initial work of genetic improvement of Chinese fir in the early 1960. With the 40 years effort for genetic improvement in Chinese fir, we have made great achievements in the establishment of seed orchard, provenance test, progeny test, hybridization, clonal forestry, joint genetic improvement for growth and wood properties etc. In order to accelerate the process of genetic improvement in Chinese fir and combine the new technique of molecular genetic marker with the methods of traditional forest genetics and breeding, we used F 1 progeny from cross J 0×F 11 as mapping population. 78 primers were screened from 1?040 random 10 base primers. The RAPD amplifications of both 78 F 1 samples and two parents were made. Of the 129 RAPD markers we obtained, most of them were fit for 1∶1 segregation ratio. The distortion segregation ratio was 14.7%. The first molecular genetic linkage frame map of Chinese fir was constructed. The J 0 parent identified 8 linkage groups covering 595.2?cM, the F 11 parent with 4 linkage groups covering 315.3?cM. This layed a solid basis for further molecular genetics research of Chinese fir.
