Studies on the Methods of Production and Extraction of Toxins from Lecanosticta acicola Causing Brown Spot Needle Blight of Pine

Abstract: Brown spot needle blight of pine is an important pine di sease in south China.It is caused by Lecanosticta acicola.The pathogenic fun gi could produce toxic substances in suitable media. In this study,several media were used for toxin production and some organic solvents were used to extract t oxins from the metabolites of the pathogen in MS medium.The results indicated:PD and MS culture liquids were better media for toxin production;adding water extr acting substances of lobollolly pine needles in the two media could not evidentl y affect the yields of LA-toxins;among these tested solvents,methyl,alcohol,eth anol,n-butanol and CHCl_3-MeOH mixed solvent(CHCl_3∶MeOH=2∶3)could extract LA-toxins fully;acetic ester and benzol could not extract the toxins f ully.According to the results of bioassay and HPLC the author believed using MS culture liquid to culture the pathogen and using the CHCl_3-MeOH mixed sol vent(CHCl_3∶MeOH=2∶3)to extract crude LA-toxins would be suitable to the production and seperation of LA-toxins.
