Proliferation and development of globular somatic embryos of Aesculus chinensis Bunge in suspension cultures

Abstract: The proliferation and development of globular somatic embryos under liquid cultural condition were investigated in this paper. The somatic embryonic callus of Aesculus chinensis Bunge from solid cultures were used as starting material. The results showed that the high synchronization globular somatic embryos of Aesculus chinensis could be obtained in MS basic medium supplemented with 3 mg/L 2,4D,1 mg/L NAA, 1.9 mg/L KNO3, and 30 g/L sucrose; Whereas the globular embryos could be able to maintain in the developmental stage both by MS + 2 mg/L 2,4D + 0.2 mg/L KT + 40 g/L sucrose, and MS + 3 mg/L 2,4D + 0.5 mg/L BA + 40 g/L sucrose. By contrast, the situation remained in globular somatic embryos could be stimulated to restart the developmental process with MS + 0.02 mg/L TDZ + 30 g/L sucrose, meanwhile, this medium could facilitate the production of cotyledon embryos. Two suspension systems have been established for globular somatic embryos proliferation and differentiation respectively. Meanwhile, we also tracked the change of pH value in liquid medium simultaneously. The various morphologies of a series of somatic embryos development under the two suspension cultures conditions were also observed. We proposed that the suspension culture of globular somatic embryos could contribute greatly to optimize the system of rapid micropropagation technique of Aesculus chinensis Bunge.
