Characteristics of cellulase inducement and hydrolysis by steam exploded corn stover

Abstract: The characteristics of cellulase inducement and hydrolysis by steam exploded corn stover were investigated. Steam explosion could remove hemicellulose from corn stover and the acquired residue was a good inducer for cellulase biosynthesis using Trichoderma reesei NL02. When the concentration of substrate was 10 g/L, the filter paper activity reached 1.90 U/mL after cultivation for seven days. The four produced and the two other enzymes were characterized by various enzyme activity measurements and steam exploded corn stover was hydrolyzed by them. The result showed that the highest sugar yields were obtained with the enzyme induced by steam exploded corn stove after cultivating for five days. When 50 g/L steam exploded residue was treated with the enzyme dosage of 25 U/g, the enzymatic hydrolysis yield was 95.9% for 24 h and the glucose yield was 76.4% for 48 h, and when the substrate concentration was improved to 80 g/L with the dosage of 15 U/g, the hydrolysis yield was 90.1% and the glucose yield was 62.9% for 48 h under the same conditions.
