Abstract: Using stem tip of Clematis ‘MultiBlue’ as explants and 1/2 MS as culture media, the suitable conditions for induction of embryogenic callus and somatic embryos, as well as plant regeneration, were determined. The results indicated that the optimal conditions for induction of embryogenic callus were 1/2 MS supplemented with 1.0 mg/L TDZ, 0.5 mg/L NAA and 2 % scourse plus 0.7 % agar. Explants cultured in this solid medium could form yellow massive or granular callus and produced embryogenic callus after subculture. Suitable media for somatic embryo induction were 1/2 MS added with 0.2 mg/L TDZ and 0.1 mg/L IBA plus 0.5 g/L CH, with an induction rate of 96%. The germination rate of somatic embryos was the highest when the plant growth regulators were 0.2 mg/L 6BA and 0.05 mg/L IBA. The plantlets with root were successfully transplanted to the medium with vermiculite. The survival rate of regeneration plant could be up to at least 80 %.
