Mitosis and chromosome number in poplar pathogenic fungus Marssonina brunnea

Abstract: Mitotic division of Marssonina brunnea was analyzed by light microscopy and the germ tube burst method (GTBM). The mitosis of M. brunnea has four phases. The chromosomes gradually shortened and thickened during prophase, and distinctly visible at metaphase. In anaphase, the chromatids separated and moved to opposite poles, at last, daughter nuclei were shaped in telophase. Three kinds of dyes were utilized to stain the chromosomes of M. brunnea, they were: DAPI, DAPI/PI and Giemsa. We found that Giemsa staining performed best that chromosome number was easily counted at the metaphase stage. Chromosomes were morphologically characterized by absolute and relative sizes.
