Cloning, expression and interaction analysis of MAGPIE and JACKDAW genes in Populus

Abstract: The PeMGP and PeJDK genes were cloned from adventitious roots of Populus, and their full-length cDNA sequences contain open reading frame(ORF)of 1 434 bp and 1 518 bp, respectively. There were two introns in their genomic structure. The two deduced proteins all contained 4 identical zinc finger domains, ZF1(CX2-4CX3FX4LX2HX3-5H)and ZF2(CX4CXnHXnX4H), ZF3(CX2CXnHX3C)and ZF4(CXCXnHX3C). Quantitative real-time PCR analysis revealed that two genes expressed in different organs, and showed a dynamic expression pattern in Populus adventitious roots. Transient gene expression assay in Populus mesophyll protoplasts demonstrated that PeMGP and PeJDK were localized in nucleus, and their protein interact each other in nucleus.
