Isolation and purification of an odorant binding protein from Batocera lineolata Chevrolat

Abstract: The fluorescence binding assays and Sephadex G75 chromatography were conducted to investigate the recognition of odorants by Batocera lineolata Chevrolat. Additionally, one of the odorant binding proteins in B. lineolata was isolated and purified. The results showed that the odorant binding protein of B. lineolata could combine with fluorescent probe(N-phenyl-1-naphthylamine, 1-NPN). Besides, twelve kinds of macromolecular crude extract protein(pro. Ⅰ-Ⅻ, Protein Ⅰ-Ⅻ)were found in antenna of B. lineolata. When the 12 kinds of macromolecular crude extract protein were isolated and purified by Sephadex G75, a purified protein(pur-pro. I, purified protein I)was found, and it could also combine with fluorescent probe(N-phenyl-1-naphthylamine, 1-NPN). The relative molecular weight of pur-pro. I was approximately 15.2 ku, and it was pro. Ⅻ(Protein Ⅻ), when contrasted with all kinds of macromolecular crude extract protein. All above, it declared that the isolated and purified protein pur-pro. I was one of the odorant binding proteins in B. lineolata.
