Study of impact factor on the anther culture in vitro of Camellia oleifera

Abstract: The anther of Camellia yuhsiensis were selected as material. The effects of different culture conditions, including different developing period of pollen, time of low temperature pretreatment, and kinds of additives, on inducing callus were investigated. Then we further studied the influence of different plant growth regulators and ratio on callus proliferation. As a result, the best developing stage of pollen on inducing callus from oiltea anther is tetrad stage; the time of low temperature pretreatment should be below 10 days; and the best medium additives is AgNO3, with the high induction rate of 8102 %. In the culture of callus proliferation, the best combination is 6BA and NAA, and the best recipe of callus proliferation culture medium is MS+6BA(20 mg/L)+NAA(005 mg/L).
