Effect of dimethyl sulphate on phytoplasma of Paulownia tomentosa seedling infected by witches’ broom and its DNA loci at SSR level

Abstract: The effect of different concentration of dimethyl sulphate(DMS)on the morphological and phytoplasma changes of Paulownia tomentosa seedlings infected by witches’ broom and DNA base sequences changes at SSR level were investigated by morphological observation, net-PCR and SSR analysis. The results indicated that the seedlings infected by witches’ broom which were treated with optimal concentration DMS might be cured, however there were still exist phytoplasma after treated with 15 mg/L DMS for 5 h. The seedlings were not alive when DMS concentration was larger than 150 mg/L. Moreover, the DNA loci at SSR level of the healthy, infected and the infected-treated seedlings were the same.
