Selection of an antagonistic bacterium against Arthrinium phaeospermum and its antibacterial protein analysis

Abstract: An antagonistic bacterial strain YB-15 against Arthrinium phaeospermum was isolated from Bambusa pervariabilis×Dendrocalamopsis grandis. The strain was identified as Bacillus velezensis by the morphological, physiological and biochemical characteristics, and 16SrDNA sequence analysis. One of the antibacterial protein was purified by using ammonium sulfate precipitation, the Sephadex G-50 chromatography, High Q Sepharose Fast Flow chromatography and Sephadex G-75 chromatography.The molecular weight of the antibacterial protein was estimated to be 31.0 ku by SDS-PAGE analysis.Stability test showed that the antibacterial protein was resistant to proteinase K and trypsin,thermal stable, sensitive to pH and ultraviolet, had different sensitivity to organic solvents.
