Rapid identification of Carpomya vesuviana Costa and five relative species of fruit flies by PCR-RFLP

Abstract: Carpomya vesuviana Costa has been a new quarantine pest in China. In this paper, the method of rapid identification of C. vesuviana Costa and 5 relative species of fruit flies was studied based on PCR-RFLP of the mitochondrial DNA(mtDNA). The results showed that DNAs, which were estimated about 350 bp and 450 bp respectively, were obtained by means of PCR with two pairs of primers designed from mtDNA of target pests.The PCR-products could be registered by restriction enzymes(Dra I and Mse I). All the 6 species could be discriminated according to the banding patterns of the restriction enzymes. It is suggested that the method is reliable for quick discrimination of fruit flies.
