Callus induction and plant regeneration from zygotic embryos of Cryptomeria fortunei

Abstract: In this article,immature seeds from 4 families were selected to conduct callus induction and then regeneration plants were obtained by organogenesis in Cryptomeria fortunei by orthogonal design with different types of hormones and concentration. The effect caused by respective factors and interactions on callus induction from zygotic embryos of Cryptomeria fortunei were studied. The following were the results: ① The amount of callus induction from 4 families increased obviously collecting seeds in the mid August. By the late August, the callus induction rate from each zygotic embryos reached the highest, among which family NO. 57 family took the first place(92%). ② There were certain differences between the induction rate by using different hormone combinations. The highest induction rate of callus was that of family NO. 57 family which grew on DCR+6-BA 1.0 mg/L +2,4-D 1.0 mg/L culture medium,and the callus induced was white, dry and of highly dense form, with no needle. ③ Regeneration plants were successfully abtained from callus on the DCR basic medium.
