A Study on Detection Technique of Bursaphelenchus xylophilus and B. mucronatus by RAPD

Abstract: By RAPD technique,primer OPK09 and pairwise primers OPC18 and OPN18 were screened from 140 arbitrary oligonuleotide primers. A specific about 2 400 bp fragment was obtained in the amplified products of 12 strains of Bursaphelenchus xylophilus using primer OPK09,and another approximate 970 bp specific fragment was amplified from 10 strains of B. mucronatus using pairwise primers OPC18 and OPN18. Experiment indicated that primer OPK09 and pairwise primers OPC18 and OPN18 have high specificity and sensitivity to all strains of B. xylophilus and B. mucronatus. So B. xylophilus and B. mucronatus can be identified rapidly and accurately using these two groups of primers.
