Improvement of Plant Cryo-sectioning Technique

Abstract: It was a time consuming process if technique of paraffin section was used to observe plant specimen because fussy processes such as dehydrating, infiltrating and embedding were needed before being observed. Cryo-sectioning technique was used to observe leafstalk, leaf, stem and root structure of Schisandra chinensis (Turez.)Ball.. The operating process was simple and efficient. Clear image could be acquired within one day. The crucial step were to select a proper concentration of glycerol, a suitable temperature of freezing and an appropriate way of section-flatting.
