Study on cryopreservation of auxiliary buds of resistant Pinus densiflora

Abstract: The conditions for cryopreservation were studied at storage temperature, storage time, sorbitol contention and the combination of 6BA and NAA levels for better understanding of reservation and effective utilization of resistant Pinus densiflora. The results showed that, when reserved at 17 ℃, survival rate of auxiliary buds could be up to 96.7 %, sorbitol and paclobutrazol could alleviate chilling injury, 0.2 mg/L 6BA+(0.1—0.2) mg/L NAA was the best combination of cryopreservation, which could improve the multiplication coefficient to 5.9—6.0 in the course of recovery growth, and the effects of cryopreservation were greatly different from each other,1#-A, 7#-7,16#-2 were elite genotype. It also provided reference for the reservation of other germplasm resources.
