Progress of wood identification based on DNA methods

Abstract

From wood identification points of view, DNA extraction methods(CTAB, SDS, PTB, DNeasy Plant Mini Kit etc.)and DNA barcode and fingerprint and wood identification methods based on DNA were summarized. For the DNA barcode concerned, the emphases were on the signature sequences in genome DNA(chloroplast DNA, cpDNArbcL,matK,trnH-psbAintergenic spacer sequences,and ribosomal DNA, rDNA ITS)as well as identification form(variable sites and phylogenetic tree)described. For the DNA fingerprint method,the emphases were on the progress and outlook of 4 kinds of molecular makers such as random amplificated polymorphism DNA, RAPD,inter-simple sequence repeat, ISSR、simple sequence repeats, SSR,single nucleotide polymorphism, SNP. It was concluded that wood identification based on DNA methods is feasible in theory but needs more studied on.
